This document specifies the quantitative liquid chromatographic determination of specific sugars (galactose, glucose, fructose, sucrose, lactose and maltose) in various milk and milk products, applying arabinose as an internal standard. The method is applicable to the following dairy matrices: milk, sweetened condensed milk, milk powder, cheese, whey powder, infant formula, milk dessert and yoghurt. The method does not apply to dairy products containing soy or to the determination of the lactose content in low-lactose milk products at levels below 1 mg/g. A high performance anion exchange chromatography method in combination with pulsed amperometric detection (HPAEC-PAD) method is applied[5][3][4]. With this method, thirteen different monosaccharides, disaccharides and trisaccharides can be separated: fucose, arabinose, galactose, glucose, fructose, sucrose, lactose, lactulose, maltose, melibiose, trehalose, isomaltulose and maltotriose. The method is applicable to labelling for the six most important sugars that can be present by nature or by addition in milk and milk products. The method does not apply to sugar contents less than 0,1 %.

Le présent document spécifie la détermination quantitative par chromatographie en phase liquide de certains sucres (galactose, glucose, fructose, saccharose, lactose et maltose) dans divers laits et produits laitiers, en utilisant l'arabinose comme étalon interne. Cette méthode s'applique aux matrices laitières suivantes: lait, lait concentré sucré, lait en poudre, fromage, lactosérum en poudre, formule infantile, dessert lacté et yaourt. Cette méthode ne s'applique pas aux produits laitiers contenant du soja ni à la détermination de la teneur en lactose des produits laitiers à faible teneur en lactose inférieure à 0,1 mg/g. Cette méthode s'appuie sur la chromatographie d'échange d'anions à haute performance couplée à la détection par ampérométrie pulsée (HPAEC-PAD)[5][3][4]. 1 Scope
This document specifies the quantitative liquid chromatographic determination of specific sugars

4 Principle
The sugars present in the sample are extracted with an aqueous ethanol buffer solution in order to
inhibit potential probiotic activities. The obtained extract is deproteinized with a Carrez clarification.
After clarification, the solution is diluted and the sugars present are separated and quantified by
HPAEC. HPAE allows carbohydrates separation at high pH. In order to improve sensitivity and stability,
post-column sodium hydroxide solution is added to the HPAEC-PAD. GOS (galacto-oligosaccharides) and
[5]
fructans do not interfere with the analysis of the sugars . Arabinose is applied as an internal standard
for the quantification of the sugars.

5 Reagents
Use only reagents of recognized analytical grade and water in accordance with ISO 3696, unless
otherwise specified.
5.1 Water, conforming to ISO 3696, grade 3 and grade 1.
5.2 Sodium hydroxide (NaOH) pellets.
5.3 Aqueous sodium hydroxide solution, substance concentration c = 1 mol/l.
Add to a 1 000 ml volumetric flask 40 g ± 1 g NaOH pellets (5.2), dissolve in about 500 ml of water and,
after cooling down, dilute with water to the mark and homogenize.
5.4 Sodium hydroxide solution, mass fraction w(NaOH) = 33 % in water.
5.5 Sodium hydroxide solution, mass fraction w(NaOH) = 50 % in water.
The amount of carbonate and mercury in the reagent should be minimized. Do not shake or stir the
solution before use. A suitable commercially available carbonate sodium hydroxide solution may also
be used.
5.6 Concentrated hydrochloric acid (HCl), mass fraction of 36 % to 38 % in water.
5.7 Aqueous hydrochloric acid solution, c = 1 mol/l.
Add to a 1 000 ml volumetric flask (6.2) 500 ml of water followed by 83 ml of concentrated HCl (5.6)
and, after cooling down, dilute with water to the mark and homogenize.
5.8 Acetonitrile (HPLC quality).
5.9 Acetonitrile in water, a volume fraction of 5 % in water.
Add to a 1 000 ml volumetric flask (6.2) 50 ml of acetonitrile (5.8), dilute with water grade 3 to the
mark and homogenize.
5.10 Anhydrous sodium acetate (CH COONa) (HPLC quality).

5.11 Eluent 1 (E1), aqueous solution of sodium acetate (CH COONa), c = 1,0 mol/l.

Add to a 1 000 ml volumetric flask (6.2) about 800 ml of degassed water grade 1 (eluent 3, 5.13) followed
by 82,0 g sodium acetate (5.10). Then dilute the aqueous solution with degassed water (eluent 3, 5.13)
to the mark and homogenize. Store the eluent under an inert atmosphere.
5.12 Eluent 2 (E2), aqueous solution of carbonate free sodium hydroxide (NaOH), c = 0,2 mol/l.
Add to a 1 000 ml volumetric flask (6.2) about 800 ml of degassed water grade 1 (eluent 3, 5.13) and
purge for 15 min with helium. Add 16,0 g of sodium hydroxide solution (5.5). Then quickly dilute the
aqueous solution with degassed water grade 1 (eluent 3, 5.13) to the mark, immediately close the bottle
and homogenize. Store the eluent under an inert atmosphere.
5.13 Eluent 3 (E3), degassed water grade 1, stored under an inert atmosphere.

5.14 Eluent 4 (E4), aqueous solution of sodium acetate (CH COONa), c = 0,025 mol/l.

Add to a 1 000 ml volumetric flask (6.2) about 800 ml of degassed water grade 1 (eluent 3, 5.13) followed
by 2,05 g of sodium acetate (5.10). Then dilute the aqueous solution with degassed water (eluent 3,
5.13) to the mark and homogenize. Store the eluent under an inert atmosphere.
5.15 Post column reagent, aqueous solution of sodium hydroxide, c = 0,3 mol/l.
Add to a 1 000 ml volumetric flask (6.2) about 800 ml of degassed water grade 1 (eluent 3, 5.13). Purge
for 15 min with helium. Add 24,0 g of sodium hydroxide solution (5.5) and quickly fill up to the mark
with the degassed water grade 1 (eluent 3, 5.13). Immediately close the flask and homogenize. Store the
post column reagent under an inert atmosphere.
IMPORTANT — It is extremely important to remove dissolved carbon dioxide from the eluents
and post column reagent prior to use and during use to avoid fast reduction in detector
sensitivity. The eluents and post column reagent are maintained under an inert gas during use.
5.16 Mixture of a volume fraction of 95 % of ethanol (with a volume fraction of 96 % ethanol and
4 % of water) and a volume fraction of 5 % methanol.
5.17 Potassium hexacyanoferrate (II) trihydrate, K Fe(CN) ·3H O.

5.20 Carrez reagent I.
Weigh 106 g of K Fe(CN) ·3H O (5.17) in a 1 000 ml volumetric flask (6.2), dissolve in 800 ml of water

(5.1) and dilute with water grade 3 to the mark. Store the Carrez reagent I in the refrigerator.
5.21 Carrez reagent II.
Weigh 220 g of Zn(CH COO) ·2H O (5.18) in a 1 000 ml volumetric flask (6.2), dissolve in 800 ml water,

add 30 ml of glacial acetic acid (5.19) and dilute with water grade 3 to the mark. Store the Carrez reagent
II in the refrigerator.
IMPORTANT — Do not use Carrez reagent II with zinc sulfate.
5.22 Buffer solution of piperazine-N,N′-bis(2-ethanesulfonic acid) (PIPES) (c = 1,5 mol/l and
pH = 6,9).
Add 22,5 g of the PIPES buffer solution to a 100 ml conical flask and add 20 ml of sodium hydroxide
solution (5.3). Adjust the pH to pH = 6,9 with NaOH 33 % (5.4) in water. Transfer the PIPES buffer
solution quantitatively into a 50 ml calibrated tube and fill up with water grade 3 till 50 ml. The pH of
the obtained buffer solution shall be within the range of 6,8 to 7,0.
5.23 Arabinose.

5.30 Internal standard stock solution arabinose.
Weigh, to the nearest mg, approximately 7 g of arabinose (5.23) into a 50 ml volumetric flask (6.2). Add
about 30 ml of water grade 3 and dissolve the arabinose. Add 2,5 ml of acetonitrile (5.8), fill up to the
mark with water and homogenize the solution.
5.31 Sugar standard stock solution.
Weigh, to the nearest 0,1 mg, approximately 260 mg of the monosaccharides galactose (5.24), glucose
(5.25) and fructose (5.26), and approximately 400 mg of the disaccharide sucrose (5.27), lactose (5.28)
and maltose (5.29) into a 500 ml volumetric flask (6.2). Add about 200 ml of water grade 3 and dissolve
the sugars. Add 25 ml of acetonitrile (5.8), fill up to the mark with water grade 3 and homogenize the
solution.
5.32 Sugar standard solutions for calibration.
Prepare the different dilutions of the sugar calibration standards as specified in Table 1. Mix the
specified volumes of the internal standard stock solution arabinose (5.30) and sugar standard stock
solution (5.31) in a 200 ml volumetric flask, add about 50 ml of water grade 3 and homogenize. Add
10 ml of acetonitrile (5.8), fill up to the mark with water and homogenize.

6.16 Pulsed amperometric detector (PAD) with a stability of ±1 °C and an operating temperature
range of 20 °C to 35 °C.
Use pulsed amperometric detection and potential settings and waveforms recommended by the
instrument supplier. Example detector potential settings (versus Ag/AgCl reference) are given in
Table 2.

7 Sampling
It is important that the laboratory receives a sample that is truly representative and has not been
damaged or changed during transport and/or storage. Sampling is not a part of the method specified in
this document.
8 Preparation of the test sample
8.1 General
Prepare the sample in accordance with an appropriate sampling procedure. Sweetened condensed milk
requires a dedicated sample preparation, see ISO 2911 | IDF 35 and 8.2.
8.2 Sample preparation of sweetened condensed milk
8.2.1 Samples of recently manufactured products in which no appreciable separation of
components can be expected
Open the container, transfer all material adhering to the lid into the container and thoroughly mix by an
up-and-down movement of a spoon, in such a way that the top layers and contents of the lower corners
are moved and mixed. When the product is in a can, transfer the contents into a jar with a well-fitting
lid. When the product is in a collapsible tube, transfer as much as possible of the content to a jar with a
well-fitting lid, then cut open the tube, scrape out all material adhering to the interior and also transfer
this to the jar. Mix the contents of the jar as described above.
8.2.2 Samples of older products and samples in which separation of components can be
expected
Heat the sample in a water bath at about 40 °C until the sample has nearly reached this temperature.
Open the container and proceed as described in 8.2.1. When the product is in a can or tube, transfer
the contents to a jar, scrape out all material adhering to the walls (in the case of a collapsible tube, after

9 Procedure
9.1 Sample extraction and clean up
9.1.1 General
In cases where the integrator chromatography software package has the option “variable internal

standard” (e.g. in Chromeleon™ ), apply the procedure given in 9.1.2. In these cases, just one sample
extraction and a clean-up procedure with two different dilutions have to be done. Adjust the integrator
settings in the variable internal standard option for the 50-fold dilution.
For low sugar contents, apply the procedure given in 9.1.2.2. For high sugar contents, apply the
procedure given in 9.1.2.3. The limits for low and high sugar contents are given in Table 3.

In cases where the chromatographic integration software has no option for “variable internal standard”
and there is no prior knowledge about the sugar levels in the dairy sample to be investigated, it is
recommended to apply the procedure given in 9.1.2.2 for the low sugar levels. In cases where a sugar
peak is bigger than the corresponding sugar peak in the highest calibration standard, the procedure
given in 9.1.2.3 shall be applied for a correct quantitation of the sugar content.
9.1.2 Sample extraction and clean-up
9.1.2.1 General
Weigh, to the nearest 0,000 1 g, 1,0 g ± 0,1 g in a 50 ml calibrated tube (6.8).
Add 4 ml of the denatured ethanol (5.16), 0,125 ml of the arabinose internal standard solution (5.30)
and 0,5 ml of the PIPES buffer solution (5.22).
Mix well by vortexing (6.7) and let the sample suspension rest for 10 min at an ambient temperature.
Fill up the calibrated tube to the 25 ml mark with demineralized water grade 3 (5.1) with a temperature
of 40 °C ± 2 °C, followed by homogenization with a homogenizer (6.6) for 1 min.
NOTE Already well-dissolved samples can be homogenized by shaking.
Check the pH of the homogenized sample solution. The pH shall be within the values pH = 5,0 to pH = 7,0.
If necessary, adjust the pH with NaOH solution (5.3) or HCl solution (5.7).
Add 0,5 ml of the Carrez reagent I solution (5.20), mix and then add 0,5 ml of the Carrez reagent II
solution (5.21). Add 2,5 ml acetonitrile (5.8) and fill up with water (5.1) to the 50 ml mark. Close the
calibrated tube with the screw cap and mix well.
Filter the suspension over black band filter paper (6.4) and collect the filtrate.

1 Domaine d’application
Le présent document spécifie la détermination quantitative par chromatographie en phase liquide
de certains sucres (galactose, glucose, fructose, saccharose, lactose et maltose) dans divers laits et
produits laitiers, en utilisant l’arabinose comme étalon interne.
Cette méthode s’applique aux matrices laitières suivantes: lait, lait concentré sucré, lait en poudre,
fromage, lactosérum en poudre, formule infantile, dessert lacté et yaourt.
Cette méthode ne s’applique pas aux produits laitiers contenant du soja ni à la détermination de la
teneur en lactose des produits laitiers à faible teneur en lactose inférieure à 0,1 mg/g.
Cette méthode s’appuie sur la chromatographie d’échange d’anions à haute performance couplée à la
[5][3][4]
détection par ampérométrie pulsée (HPAEC-PAD) . Treize monosaccharides, disaccharides et
trisaccharides différents peuvent être séparés par cette méthode: fucose, arabinose, galactose, glucose,
fructose, saccharose, lactose, lactulose, maltose, mélibiose, tréhalose, isomaltulose et maltotriose.
Cette méthode s’applique à l’étiquetage des six principaux sucres qui peuvent être présents
naturellement ou ajoutés dans le lait et les produits laitiers. Cette méthode ne s’applique pas aux teneurs
en sucre inférieures à 0,1 %.

4 Principe
Les sucres présents dans l’échantillon sont extraits à l’aide d’une solution tampon aqueuse contenant de
l’éthanol afin d’inhiber d’éventuelles activités probiotiques. L’extrait ainsi obtenu est déprotéinisé par
clarification de Carrez. Après clarification, la solution est diluée et les sucres présents sont séparés et
quantifiés par HPAEC. HPAE permet la séparation des glucides à pH élevé. Afin d’améliorer la sensibilité

et la stabilité, une solution d’hydroxyde de sodium est ajoutée en post-colonne de l’HPAEC. Les GOS
[5]
(galacto-oligosaccharide) et les fructanes n’interfèrent pas avec l’analyse des sucres . L’arabinose est
utilisé comme étalon interne pour la quantification des sucres.
5 Réactifs
Utiliser uniquement des réactifs de qualité analytique reconnue et de l’eau conformément à l’ISO 3696,
sauf indication contraire.
5.1 Eau, conformément à l’ISO 3696, grade 3 et grade 1.
5.2 Pastilles d’hydroxyde de sodium (NaOH).
5.3 Solution aqueuse d’hydroxyde de sodium, de concentration c = 1 mol/l.
Introduire 40 g ± 1 g de pastilles de NaOH (5.2) dans une fiole jaugée de 1 000 ml, les dissoudre dans
environ 500 ml d’eau et, après refroidissement, diluer avec de l’eau jusqu’au repère et homogénéiser.
5.4 Solution d’hydroxyde de sodium, de fraction massique w(NaOH) = 33 % dans l’eau.
5.5 Solution d’hydroxyde de sodium, de fraction massique w(NaOH) = 50 % dans l’eau.
Il convient que le réactif contienne le moins possible de carbonate et de mercure. Ne pas agiter
ou mélanger la solution avant utilisation. Une solution d’hydroxycarbonate de sodium appropriée
disponible dans le commerce peut également être utilisée.
5.6 Acide chlorhydrique concentré (HCl), fraction massique de 36 % à 38 % dans l’eau.
5.7 Solution aqueuse d’acide chlorhydrique, c = 1 mol/l.
Introduire 500 ml d’eau et 83 ml d’HCl concentré (5.6) dans une fiole jaugée de 1 000 ml (6.2) et, après
refroidissement, diluer avec de l’eau jusqu’au repère et homogénéiser.
5.8 Acétonitrile (qualité HPLC).
5.9 Acétonitrile dans l’eau, à une fraction volumique de 5 % dans l’eau.
Introduire 50 ml d’acétonitrile (5.8) dans une fiole jaugée de 1 000 ml (6.2), diluer avec de l’eau de
grade 3 jusqu’au repère et homogénéiser.
5.10 Acétate de sodium anhydre (CH COONa) (qualité HPLC).

5.11 Éluant 1 (E1), solution aqueuse d’acétate de sodium (CH COONa), c = 1,0 mol/l.

Introduire environ 800 ml d’eau dégazée de grade 1 (éluant 3, 5.13) et 82,0 g d’acétate de sodium
(5.10) dans une fiole jaugée de 1 000 ml (6.2). Diluer ensuite la solution aqueuse à l’aide d’eau dégazée
(éluant 3, 5.13) jusqu’au repère et homogénéiser. Conserver l’éluant sous atmosphère inerte.
5.12 Éluant 2 (E2), solution aqueuse d’hydroxyde de sodium exempte de carbonate (NaOH),
c = 0,2 mol/l.
Introduire environ 800 ml d’eau dégazée de grade 1 (éluant 3, 5.13) dans une fiole jaugée de 1 000 ml (6.2)
et purger avec de l’hélium pendant 15 min. Ajouter 16,0 g de solution d’hydroxyde de sodium (5.5).
Diluer ensuite rapidement la solution aqueuse à l’aide d’eau dégazée de grade 1 (éluant 3, 5.13) jusqu’au
repère, fermer immédiatement la fiole et homogénéiser. Conserver l’éluant sous atmosphère inerte.

5.13 Éluant 3 (E3), eau dégazée de grade 1, conservée sous atmosphère inerte.
5.14 Éluant 4 (E4), solution aqueuse d’acétate de sodium (CH COONa), c = 0,025 mol/l.

Introduire environ 800 ml d’eau dégazée de grade 1 (éluant 3, 5.13) et 2,05 g d’acétate de sodium
(5.10) dans une fiole jaugée de 1 000 ml (6.2). Diluer ensuite la solution aqueuse à l’aide d’eau dégazée
(éluant 3, 5.13) jusqu’au repère et homogénéiser. Conserver l’éluant sous atmosphère inerte.
5.15 Réactif post-colonne, solution aqueuse d’hydroxyde de sodium, c = 0,3 mol/l.
Introduire environ 800 ml d’eau dégazée de grade 1 (éluant 3, 5.13) dans une fiole jaugée de 1 000 ml (6.2).
Purger avec de l’hélium pendant 15 min. Ajouter 24,0 g de solution d’hydroxyde de sodium (5.5)
et compléter rapidement jusqu’au repère avec de l’eau dégazée de grade 1 (éluant 3, 5.13). Fermer
immédiatement la fiole et homogénéiser. Conserver le réactif post-colonne sous atmosphère inerte.
IMPORTANT — Il est extrêmement important d’éliminer le dioxyde de carbone dissous dans
les éluants et le réactif post-colonne avant et en cours d’utilisation pour éviter une réduction
rapide de la sensibilité du détecteur. Les éluants et le réactif post-colonne sont maintenus dans
un environnement de gaz inerte en cours d’utilisation.
5.16 Mélange d’une fraction volumique de 95 % d’éthanol (avec une fraction volumique de 96 %
d’éthanol et de 4 % d’eau) et d’une fraction volumique de 5 % de méthanol.
5.17 Hexacyanoferrate (II) de potassium trihydraté, K Fe(CN) ·3H O.

réfrigérateur.
5.21 Réactif de Carrez II.
Introduire 220 g de Zn(CH COO) .2H O (5.18) dans une fiole jaugée de 1 000 ml (6.2), dissoudre à l’aide

de 800 ml d’eau, ajouter 30 ml d’acide acétique glacial (5.19), et diluer à l’eau de grade 3 jusqu’au repère.
Conserver le réactif de Carrez II au réfrigérateur.
IMPORTANT — Ne pas utiliser le réactif de Carrez II en présence de sulfate de zinc.
5.22 Solution tampon d’acide pipérazine-N,N′-bis(2-éthanesulfonique) (PIPES) (c = 1,5 mol/l
et pH = 6,9).
Introduire 22,5 g de solution tampon de PIPES dans une fiole conique de 100 ml et ajouter 20 ml de
solution d’hydroxyde de sodium (5.3). Ajuster le pH à pH = 6,9 à l’aide de NaOH à 33 % (5.4) dans l’eau.
Transférer quantitativement la solution tampon de PIPES dans un tube étalonné de 50 ml et compléter
avec de l’eau de grade 3 jusqu’à atteindre 50 ml. Le pH de la solution tampon ainsi obtenue doit être
compris entre 6,8 et 7,0.

Peser, au mg près, environ 7 g d’arabinose (5.23) dans une fiole jaugée de 50 ml (6.2). Ajouter
environ 30 ml d’eau de grade 3 et dissoudre l’arabinose. Ajouter 2,5 ml d’acétonitrile (5.8), compléter
jusqu’au repère avec de l’eau et homogénéiser la solution.
5.31 Solution mère d’étalon de sucre.
Peser, à 0,1 mg près, environ 260 mg des monosaccharides galactose (5.24), glucose (5.25) et
fructose (5.26) et environ 400 mg des disaccharides saccharose (5.27), lactose (5.28) et maltose (5.29)
dans une fiole jaugée de 500 ml (6.2). Ajouter environ 200 ml d’eau de grade 3 et dissoudre les sucres.
Ajouter 25 ml d’acétonitrile (5.8), compléter jusqu’au repère avec de l’eau de grade 3, et homogénéiser
la solution.
5.32 Solutions étalons de sucre pour l’étalonnage.
Préparer les différentes dilutions des solutions d’étalonnage de sucre indiquées dans le Tableau 1.
Mélanger les volumes indiqués de solution mère d’étalon interne d’arabinose (5.30) et de solution
mère d’étalon de sucre (5.31) dans une fiole jaugée de 200 ml, ajouter environ 50 ml d’eau de grade 3
et homogénéiser. Ajouter 10 ml d’acétonitrile (5.8), compléter jusqu’au repère avec de l’eau et
homogénéiser.

7 Échantillonnage

8 Préparation de l’échantillon pour essai
8.1 Généralités
Préparer l’échantillon conformément à un mode opératoire d’échantillonnage approprié. Le lait
concentré sucré implique une préparation de l’échantillon dédiée (voir l’ISO 2911 | FIL 35 et en 8.2).
8.2 Préparation des échantillons de lait concentré sucré
8.2.1 Échantillons de produits fabriqués récemment dans lesquels aucune séparation
appréciable des constituants ne peut être attendue
Ouvrir le récipient, transférer la totalité du produit adhérant au couvercle dans le récipient et bien
homogénéiser le tout en déplaçant verticalement une cuillère, de façon à déplacer et à mélanger les
couches supérieures ainsi que le contenu des coins inférieurs. Lorsque le produit se trouve dans un
récipient en métal, transférer son contenu dans un bocal muni d’un couvercle bien ajusté. Lorsque le
produit se trouve dans un tube souple, transférer le maximum de son contenu dans un bocal muni d’un
couvercle bien ajusté, et ouvrir le tube en le découpant, gratter l’ensemble du produit adhérant aux
parois internes et le transférer également dans le bocal. Mélanger le contenu du bocal comme indiqué
ci-dessus.

Chauffer l’échantillon au bain-marie à environ 40 °C jusqu’à ce que l’échantillon ait quasiment atteint

Milk and milk products — Determination of the
sugar contents — High performance anion exchange
chromatographic method (HPAEC-PAD)
1 Scope
The document specifies the quantitative liquid chromatographic determination of specific sugars
(galactose, glucose, fructose, sucrose, lactose, and maltose) in various milk and milk products, applying
arabinose as internal standards. The method is applicable for the following different dairy matrices:
milk, milk powder, cheese, whey powder, infant formula, milk dessert and yogurt.
Soy containing dairy products are excluded. The determination of the lactose content in low lactose
milk products is excluded.
High performance anion exchange chromatographic method in combination with pulsed amperometric
detection (HPAEC-PAD) is applied.
With this method the following thirteen different mono- and disaccharides can be separated: fucose,
arabinose, galactose, glucose, fructose, sucrose, lactose, lactulose, maltose, melibiose, trehalose,

isomaltulose (e.g. palatinose ) and maltotriose.
The method is especially meant for labelling purposes of the six most important sugars which can be
present by nature or by addition in milk and milk products. The method is not applicable for sugar
contents less than 0,1%.
2 Normative references

EN ISO 3696, Water for analytical laboratory use — Specification and test methods
3 Principle
The sugars present in the sample are extracted with an aqueous ethanol buffer solution in order to
inhibit possible probiotic activities. The obtained extract is deproteinized with Carrez clarification.
After clarification the solution is diluted and the sugars present are separated and quantified by high
performance anion exchange chromatography (HPAEC). In order to improve the sensitivity and stability
of the pulsed amperometric detector (PAD), post-column a sodium hydroxide solution was added to the
HPAEC effluent with pulsed amperometric detection (HPAEC-PAD). GOS and fructans don’t interfere.
Arabinose is applied as internal standard for the quantitation of the sugars.
4 Reagents
Use only reagents of recognized analytical grade and water according to EN ISO 3696, unless otherwise
specified.
4.1 Water, complying with EN ISO 3696, grade 3.
4.2 Sodium hydroxide (NaOH) pellets.

4.3 Aqueous sodium hydroxide solution, substance concentration c = 1 mol/l
Add to a 1 000 ml volumetric flask 40 g ± 1 g NaOH pellets (4.2), dissolve in about 500 ml of water and
after cooling down, dilute with water to the mark and homogenize.
4.4 Aqueous sodium hydroxide solution, c = 4 mol/l
Add to a 1 000 ml volumetric flask 160 g ± 1 g NaOH pellets (4.2), dissolve in about 500 ml of water and
after cooling down, dilute with water to the mark and homogenize.
4.5 Sodium hydroxide solution, mass fraction w(NaOH) = 33 % in water.
4.6 Sodium hydroxide solution, w(NaOH) = 50 % in water.
The reagent should contain the minimum amount of carbonate and mercury. Do not shake or stir the
solution before use.
4.7 Concentrated hydrochloric acid, 36 % to 38 %
4.8 Aqueous hydrochloric acid solution, c = 1 mol/l
Add to a 1 000 ml volumetric flask (5.2) 500 ml of water followed by 83 ml of concentrated HCl (4.7) and
after cooling down, dilute with water to the mark and homogenize.
4.9 Acetonitrile
4.10 Acetonitrile in water, 5 % in water
Add to a 1 000 ml volumetric flask (5.2) 50 ml of acetonitrile (4.9), dilute with water to the mark and
homogenize.
4.11 Anhydrous sodium acetate (CH COONa)

4.12 Eluent 1 (E1), aqueous solution of sodium acetate (CH COONa), c = 1,0 mol/l.

Add to a 1 000 ml volumetric flask (5.2) about 800 ml of degassed water (eluent 3, 4.14) followed by
82 g sodium acetate (4.11). Then dilute the aqueous solution with degassed water (eluent 3, 4.14) to the
mark and homogenize. Store the eluent under inert atmosphere.
4.13 Eluent 2 (E2), aqueous solution of carbonate free sodium hydroxide (NaOH), c = 0,2 mol/l
Add to a 1 000 ml volumetric flask (5.2) about 800 ml of degassed water (eluent 3, 4.14) and purge for
15 min. with helium. Add 16,0 g of sodium hydroxide solution (4.6). Then quickly dilute the aqueous
solution with degassed water (eluent 3, 4.14) to the mark, close immediately the bottle and homogenize.
Store the eluent under inert atmosphere.
4.14 Eluent 3 (E3), degassed water, stored under inert atmosphere.
4.15 Eluent 4 (E4), aqueous solution of sodium acetate (CH COONa), c = 0,025 mol/l

Add to a 1 000 ml volumetric flask (5.2) about 800 ml of degassed water (eluent 3, 4.14) followed by
2.05 g of sodium acetate (4.11). Then dilute the aqueous solution with degassed water (eluent 3, 4.14) to
the mark and homogenize. Store the eluent under inert atmosphere.

4.16 Post column reagent, aqueous solution of sodium hydroxide, c = 0,3 mol/l.
Add to a 1 000 ml volumetric flask (5.2) about 800 ml of degassed water (eluent 3, 4.14). Purge for 15
min. with helium. Add 24 g of sodium hydroxide solution (4.6) and quickly fill up to the mark with the
degassed water (eluent 3, 4.14), close immediately the flask and homogenize. Store the post column
reagent under inert atmosphere.
Important — It is extremely important to remove dissolved carbon dioxide from the eluens
and post column reagent prior to use and during use. The eluents and post column reagent are
maintained under inert gas during use.
4.17 Mixture of ethanol 96 % (v/v) and 5 % (v/v) methanol

4.22 Carrez II reagent
Weigh 220 g of Zn(CH COO) .2H O (4.19) in a 1 000 ml volumetric flask (5.2), dissolve in 800 ml water,

add 30 ml of glacial acetic acid (4.20) and dilute with water to the mark. Store the Carrez II reagent in
the refrigerator.
IMPORTANT — Do not use Carrez II reagent with zinc sulfate.
4.23 Buffer solution of piperazine-N,N'-bis(2-ethanesulfonic acid) (PIPES) (c = 1,5 mol/l and
pH = 6,9)
Add 22,5 g of PIPES to an 100 ml Erlenmeyer flask and dissolve in 20 ml of sodium hydroxide solution
(4.3). Adjust the pH to pH = 6,9 with NaOH 33 % (4.5) in water. Transfer the PIPES buffer solution
quantitatively into a 50 ml calibrated tube and fill up with demineralized water till 50 ml. The pH of the
obtained buffer solution shall be within the range of 6,8 to 7,0.

4.31 Internal standard stock solution arabinose
Weigh to the nearest mg, approximately 7 g of arabinose (4.24) into a 50 ml volumetric flask (5.2). Add
about 30 ml of water and dissolve the arabinose. Add 2,5 ml of acetonitrile (4.9), fill up to the mark with
water and homogenize the solution.
4.32 Sugar standard stock solutions
Weigh to the nearest 0,1 mg, approximately 260 mg of the monosaccharide galactose (4.25), glucose
(4.26) and fructose (4.27) and approximately 400 mg of the disaccharides sucrose (4.28), lactose (4.29),
and maltose (4.30) into a 500 ml volumetric flask (5.2). Add about 200 ml of water and dissolve the
sugars. Add 25 ml of acetonitrile (4.9), fill up to the mark with water and homogenize the solution.
4.33 Sugar standard solutions for calibration
Prepare the different dilutions of the sugar calibration standards as specified in Table 1. Dilute the
specified volumes of the internal standard stock solution arabinose (4.31) and sugar standard stock
solution (4.32) in a 200 ml volumetric flask, add about 50 ml of water and homogenize. Add 10 ml of
acetonitrile (4.9), fill up to the mark with water and homogenize.

6 Sampling
It is important that the laboratory receives a sample which is truly representative and has not been
damaged or changed during transport and/or storage. Sampling is not a part of the method specified in
this Standard.
7 Preparation of the test sample
7.1 General
Prepare the sample in accordance with an appropriate sampling procedure. Sweetend condensed milk
[[4]]
requires a dedicated sample preparation , see 7.2.
7.2 Sample preparation of sweetened condensed milk
7.2.1 Samples of recently manufactured products in which no appreciable separation of
components may be expected
Open the container, transfer all material adhering to the lid into the container and thoroughly mix
by an up-and-down movement of a spoon, in such a way that the top layers and contents of the lower
corners are moved and mixed. When the product is in a can, transfer the contents into a jar with a
well-fitting lid. When the product is in a collapsible tube, transfer as much as possible of the content to
a jar with a well-fitting lid, then cut open the tube, scrape out all material adhering to the interior and
transfer this also to the jar. Mix the conents of the jar as described above.
7.2.2 Samples of older products and samples in which separation of components may be
expected
Heat in a water bath at about 40°C until the sample has nearly reached this temperature. Open the
container and proceed as described in 7.2.1. When the product is in a can or tube, transfer the contents
to a jar, scrape out all material adhering to the walls (in case of a collapsible tube, after cutting open the
tube) and continue the mixing until the whole mass is homogeneous, reducing the size of any cristals by
crushing with a glass rod. Close the jar with a well-fitting lid. Allow to cool.
8 Procedure
8.1 Sample extraction and clean up
8.1.1 General
In case the integrator chromatography software package does have the option “variable internal
standard” (e.g. in Chromeleon) apply the procedure 8.1.2. In that case just one sample extraction and

clean-up procedure with two different dilutions have to be done. Adjust the integrator settings for in
the variable internal standard option for the 50-fold dilution.
Apply for low sugar contents procedure 8.1.2.1 and for high sugar contents procedure 8.1.2.2. The limits
for low and high sugar contents are given in Table 3.

In case the chromatographic integration software has no option for “variable internal standard”
and there is no prior knowledge about the sugar levels in the dairy sample to be investigated, it is
recommended to start apply the protocol 8.1.2.1 for the low sugar levels. In case a sugar peak is bigger
than the corresponding sugar peak in the highest calibration standard, procedure 8.1.2.2 shall be
applied for a correct quantitation of the sugar content.
8.1.2 Sample extraction and clean-up
Weigh to the nearest 0,0001 g, approximately 1,0 g ± 0,1 g in a 50 ml calibrated tube (5.8).
Add 4 ml of the mixture of ethanol and methanol (4.17), 0,125 ml of arabinose internal standard solution
(4.31) and 0,5 ml PIPES buffer solution (4.23).
Mix well by vortexing (5.7) and let the sample suspension rest for 10 min at ambient temperature.
Fill up the calibrated tube to the 25 ml mark with demineralized water grade 3 (4.1) with a temperature
of 40 °C ± 2 °C, followed by homogenization with an ultra-turrax (5.6) during 1 min.
NOTE Already well dissolved samples can be homogenized by shaking.
Check the pH of the homogenized sample solution. The pH shall be within the values pH = 5,0 to pH = 7,0.
If necessary, adjust the pH with NaOH solution (4.3) or HCl solution (4.8).
Add with a dispenser (5.10) first 0,5 ml Carrez I reagent solution (4.21), mix, 0,5 ml Carrez II reagent
solution (4.22), add with a dispenser (5.10) 2,5 ml acetonitrile (4.9) and fill up with water (4.1) to the
50 ml mark, close the calibrated tube with the screw cap and mix well.
Filter the suspension over black band filter paper (5.4) and collect the filtrate.
Prepare from each filtrate two different dilutions directly in the HPLC vials (5.11)
— 10-fold dilution: mix 100 μl of filtrate with 900 μl of water grade 3 (4.1)
— 50-fold dilution: mix 20 μl of filtrate with 980 μl of water grade 3 (4.1) and adjust the integrator
settings for these 50-fold diluted filtrates in the variable internal standard procedure.
Inject 5 μl of the diluted filtrates in the high performance anion exchange chromatographic system (5.12).
In case of the measured sugar peak is outside the calibration graph in the 10-fold diluted sample, use
the results which are measured in the 50-fold diluted samples after proper adjusting the settings for
the variable internal standard option.

1 Domaine d’application
Le présent document spécifie la détermination quantitative par chromatographie en phase liquide de
certains sucres (galactose, glucose, fructose, saccharose, lactose et maltose) dans divers laits et produits
laitiers, en utilisant l’arabinose comme étalon interne. Cette méthode s’applique aux différentes
matrices laitières suivantes : lait, lait en poudre, fromage, lactosérum en poudre, formule infantile,
dessert lacté et yaourt.
Les produits laitiers contenant du soja sont exclus du domaine d’application du présent document. La
détermination de la teneur en lactose des produits laitiers à faible teneur en lactose est également
exclue.
Cette méthode s’appuie sur la chromatographie d’échange d’anions à haute performance couplée à la
détection par ampérométrie pulsée (HPAEC-PAD).
Les treize différents mono et disaccharides suivants peuvent être séparés par cette méthode : fucose,
arabinose, galactose, glucose, fructose, saccharose, lactose, lactulose, maltose, mélibiose, tréhalose,
TM
isomaltulose (par exemple palatinose ) et maltotriose.
Cette méthode est spécialement destinée à l’étiquetage des six principaux sucres qui peuvent être
présents naturellement ou ajoutés dans le lait et les produits laitiers. Cette méthode ne s’applique pas
aux teneurs en sucre inférieures à 0,1 %.
2 Références normatives

EN ISO 3696, Eau pour laboratoire à usage analytique — Spécification et méthodes d’essai.
3 Principe
Les sucres présents dans l’échantillon sont extraits à l’aide d’une solution tampon aqueuse contenant de
l’éthanol afin d’inhiber d’éventuelles activités probiotiques. L’extrait ainsi obtenu est déprotéinisé par
clarification de Carrez. Après clarification, la solution est diluée et les sucres présents sont séparés et
quantifiés par chromatographie d’échange d’anions à haute performance (HPAEC). Afin d’améliorer la
sensibilité et la stabilité du détecteur par ampérométrie pulsée (PAD), une solution d’hydroxyde de
sodium a été ajoutée en post-colonne à l’effluent de HPAEC avec détection par ampérométrie pulsée
(HPAEC-PAD). Les GOS et les fructanes ne provoquent pas d’interférence. L’arabinose est utilisé comme
étalon interne pour la quantification des sucres.

4 Réactifs
Utiliser uniquement des réactifs de qualité analytique reconnue et de l’eau conformément à
l’EN ISO 3696, sauf indication contraire.
4.1 Eau, conforme à l’EN ISO 3696, de grade 3.
4.2 Pastilles d’hydroxyde de sodium (NaOH)
4.3 Solution aqueuse d’hydroxyde de sodium, de concentration en substance c = 1 mol/l.
Introduire 40 g ± 1 g de pastilles de NaOH (4.2) dans une fiole jaugée de 1 000 ml, les dissoudre dans
environ 500 ml d’eau puis, après refroidissement, diluer avec de l’eau jusqu’au repère et homogénéiser.
4.4 Solution aqueuse d’hydroxyde de sodium, c = 4 mol/l.
Introduire 160 g ± 1 g de pastilles de NaOH (4.2) dans une fiole jaugée de 1 000 ml, les dissoudre dans
environ 500 ml d’eau puis, après refroidissement, diluer avec de l’eau jusqu’au repère et homogénéiser.
4.5 Solution d’hydroxyde de sodium, de fraction massique w(NaOH) = 33 % dans l’eau.
4.6 Solution d’hydroxyde de sodium, w(NaOH) = 50 % dans l’eau.
Il convient que le réactif contienne le moins possible de carbonate et de mercure. Ne pas agiter ou
mélanger la solution avant utilisation.
4.7 Acide chlorhydrique concentré, de 36 % à 38 %.
4.8 Solution aqueuse d’acide chlorhydrique, c = 1 mol/l.
Introduire 500 ml d’eau puis 83 ml d’HCl concentré (4.7) dans une fiole jaugée de 1 000 ml (5.2) puis,
après refroidissement, diluer avec de l’eau jusqu’au repère et homogénéiser.
4.9 Acétonitrile
4.10 Acétonitrile dans l’eau, à 5 % dans l’eau.
Introduire 50 ml d’acétonitrile (4.9) dans une fiole jaugée de 1 000 ml (5.2), diluer avec de l’eau
jusqu’au repère et homogénéiser.
4.11 Acétate de sodium anhydre (CH COONa).

4.12 Éluant 1 (E1), solution aqueuse d’acétate de sodium (CH COONa), c = 1,0 mol/l.

Introduire environ 800 ml d’eau dégazée (éluant 3, 4.14) puis 82 g d’acétate de sodium (4.11) dans une
fiole jaugée de 1 000 ml (5.2). Puis diluer la solution aqueuse à l’aide d’eau dégazée (éluant 3, 4.14)
jusqu’au repère et homogénéiser. Conserver l’éluant sous atmosphère inerte.
4.13 Éluant 2 (E2), solution aqueuse d’hydroxyde de sodium exempte de carbonate (NaOH),
c = 0,2 mol/l.
Introduire environ 800 ml d’eau dégazée (éluant 3, 4.14) dans une fiole jaugée de 1 000 ml (5.2) puis
purger avec de l’hélium pendant 15 min. Ajouter 16,0 g de solution d’hydroxyde de sodium (4.6). Puis
diluer rapidement la solution aqueuse à l’aide d’eau dégazée (éluant 3, 4.14) jusqu’au repère, fermer
immédiatement la fiole et homogénéiser. Conserver l’éluant sous atmosphère inerte.

4.14 Éluant 3 (E3), eau dégazée, conservée sous atmosphère inerte.

Introduire environ 800 ml d’eau dégazée (éluant 3, 4.14) puis 2,05 g d’acétate de sodium (4.11) dans
une fiole jaugée de 1 000 ml (5.2). Puis diluer la solution aqueuse à l’aide d’eau dégazée (éluant 3, 4.14)
jusqu’au repère et homogénéiser. Conserver l’éluant sous atmosphère inerte.
4.16 Réactif post-colonne, solution aqueuse d’hydroxyde de sodium, c = 0,3 mol/l.
Introduire environ 800 ml d’eau dégazée (éluant 3, 4.14) dans une fiole jaugée de 1 000 ml (5.2). Purger
avec de l’hélium pendant 15 min. Ajouter 24 g de solution d’hydroxyde de sodium (4.6) puis compléter
rapidement jusqu’au repère avec de l’eau dégazée (éluant 3, 4.14), fermer immédiatement la fiole et

4.22 Réactif de Carrez II
Introduire 220 g de Zn(CH COO) .2H O (4.19) dans une fiole jaugée de 1 000 ml (5.2), dissoudre à l’aide

de 800 ml d’eau, ajouter 30 ml d’acide acétique glacial (4.20), puis diluer à l’eau jusqu’au repère.
Conserver le réactif de Carrez II au réfrigérateur.
IMPORTANT — Ne pas utiliser le réactif de Carrez II en présence de sulfate de zinc.
4.23 Solution tampon d’acide pipérazine-N,N’-bis(2-éthanesulfonique) (PIPES) (c = 1,5 mol/l et
pH = 6,9)
Introduire 22,5 g de PIPES dans un erlenmeyer de 100 ml et les dissoudre dans 20 ml de solution
d’hydroxyde de sodium (4.3). Ajuster le pH à pH = 6,9 à l’aide de NaOH à 33 % (4.5) dans l’eau.
Transférer quantitativement la solution tampon de PIPES dans un tube étalonné de 50 ml et compléter
avec de l’eau déminéralisée jusqu’à atteindre 50 ml. Le pH de la solution tampon ainsi obtenue doit être
compris entre 6,8 et 7,0.

4.31 Solution mère d’étalon interne d’arabinose
Peser, au mg près, environ 7 g d’arabinose (4.24) dans une fiole jaugée de 50 ml (5.2). Ajouter environ
30 ml d’eau et dissoudre l’arabinose. Ajouter 2,5 ml d’acétonitrile (4.9), compléter jusqu’au repère avec
de l’eau puis homogénéiser la solution.
4.32 Solutions mères d’étalons de sucre
Peser, à 0,1 mg près, environ 260 mg des monosaccharides galactose (4.25), glucose (4.26) et fructose
(4.27) et environ 400 mg des disaccharides saccharose (4.28), lactose (4.29) et maltose (4.30) dans une
fiole jaugée de 500 ml (5.2). Ajouter environ 200 ml d’eau et dissoudre les sucres. Ajouter 25 ml
d’acétonitrile (4.9), compléter jusqu’au repère avec de l’eau puis homogénéiser la solution.
4.33 Solutions étalons de sucre pour l’étalonnage
Préparer les différentes dilutions des solutions d’étalonnage de sucre indiquées dans le Tableau 1.
Diluer les volumes indiqués de solution mère d’étalon interne d’arabinose (4.31) et de solution mère
d’étalon de sucre (4.32) dans une fiole jaugée de 200 ml, ajouter environ 50 ml d’eau et homogénéiser.
Ajouter 10 ml d’acétonitrile (4.9), compléter jusqu’au repère avec de l’eau puis homogénéiser.
